Highly selective strategy for the extraction with salting-out method of bio 1,5-pentanediol produced from Escherichia coli whole-cell reaction

1,5-Pentanediol (1,5-PDO) is a valuable C5 diol with broad industrial applications, and its biosynthesis via whole-cell catalysis has garnered interest. However, efficient recovery of biogenic 1,5-PDO from aqueous media remains underexplored. In this study, we aimed to develop a selective liquid-liquid extraction method incorporating a salting-out strategy. n-Butanol was identified as the most effective solvent, and linear solvent energy relationship analysis revealed that solvent cohesiveness predominantly influenced the extraction yield. As the similarity between 1,5-PDO and 5-hydroxyvalerate was observed to limit extraction purity, the issue was addressed by introducing a salting-out method with 200 mM potassium carbonate, which significantly improved the extraction performance. To improve economic feasibility, we demonstrated effective solvent recycling, achieving 99.9% extraction yield after four cycles. Finally, this method was further validated using a biological sample from an Escherichia coli whole-cell reaction, successfully recovering biogenic 1,5-PDO with 93.15% of extraction yield. This study successfully established a 1,5-PDO extraction method based on the liquid-liquid extraction coupled with salting-out method and demonstrated its applicability to bioprocesses.
